Outline of Annual Research Achievements

タンパク質の膜組込みは、すべての生物に保存される必須のメカニズムである。モデル生物の大腸菌においては、Secタンパク質と、YidCがタンパク質の膜組込みに関与する。主要なSecタンパク質のいくつかとYidCは，真核細胞も同様のタンパク質が存在する。タンパク質の膜組込み時に形成されるSecとYidCから構成される複合体はSecホロ複合体と呼ばれているがその実態は不明である。タンパク質の膜組込みを明らかにするためには、これらタンパク質の構造解析と、構造情報に基づく機能解析が必要である。本年度は、タンパク質の膜組込みに関連するSecタンパク質のこれまでに報告されていない形の構造をＸ線結晶構造解析で明らかとした。YidCとSecタンパク質との相互作用領域は未だ不明な点が多いが、この構造解析は今後の相互作用解析の基盤となる。Secホロ複合体は、６種類の膜タンパク質から形成される。そのうち、５種類から構成されるタンパク質複合体を新たに見出した安定変異体を用いることで、精製することに成功した。続いて結晶構造解析を進めている。構造解析だけでなく、タンパク質の膜組込みを明らかとするためには、純化されたin vitroでの実験系の確立も欠かせない。本研究期間において、新規に開発されたポリマーを用いて、効率よくナノディスクとよばれる状態に再構成する系を組んだ。膜タンパク質の基質として蛍光基をもつ特殊なタンパク質を合成し、このYidC含有ナノディスクを用いて、in vitroで膜組込みさせるべく研究を進めた。
