Outline of Annual Research Achievements

ヒトiPS細胞は、今まで入手が困難であったヒト細胞のソースとしてますます重要性が増しており、国内外において、ヒトiPS細胞由来分化細胞を薬効や安全性の創薬あるいは再生医療に利用する試みが急速に広がっている。しかしながら、分化細胞は未成熟な特徴を有するため、実用化に障壁となる可能性が考えられる。そこで、本研究において、ヒトiPS細胞由来心筋細胞を成熟化させる技術の開発を行った。まず、ヒトiPS細胞由来心筋細胞を作製して活動電位をパッチクランプ法により品質を評価したところ、自動能を有しており、最大拡張期電位(Maximum Diastolic Potential; MDP)が-50mV程度で浅めであった。次に、KCNJ2チャネルのアデノウイルスを作製しヒトiPS細胞由来心筋細胞に過剰発現させたところ、MDPが深くなり自動能が消失し、電気生理学的に成熟することを新たに見出した。さらに、この現象が株を超えて認められるのか検討を行ったところ、複数のヒトiPS/ES細胞由来心筋細胞を用いて拍動が停止したことから、成熟できる可能性が示唆された。次に、インシリコのシミュレーションを行った。ヒト心室筋モデルにKCNJ2の関与を減らすことにより、自動能を獲得して、MDPが浅い活動電位を再現することに成功した。シングルセルでhERG阻害剤であるE4031を添加したところ、30nM以上で拍動が停止したが、KCNJ2を発現させた場合には用量依存的にE4031によるQT間隔延長が認められ、インシリコモデルにより確認した。以上の結果から、成熟したiPS由来心筋細胞は創薬などへの応用が期待される。
