Muse細胞は生体内の間葉系組織に散在性に存在する自然の多能性幹細胞で腫瘍性を持たない。一方再生能の高いプラナリアも間葉系組織に散在性に存在する多能性幹細胞"neoblast"を有する。この類似性に着眼し「組織恒常性維持機構」におけるMuse細胞の機能解明を目的とする。
Muse細胞のマトリックスへの接着関連因子と多能性因子との連動を解析し、多様な組織からの単離を試みて特性解析を行い、組織恒常性維持との関連においては、劇症肝炎、脳梗塞、腎不全などのモデルを作成し、組織再生への寄与を探索した。Muse細胞は当初骨髄、皮膚、脂肪組織にあると報告され、間葉系組織にのみ存在すると想定されてきたが、本研究によって末梢血にも循環すること、また予想を超えて様々な臓器の結合組織に分布していることがわかった。特に脾臓、肝臓、肺、気管、肝臓、膵臓の結合組織にもあり、脳では脳膜、くも膜に局在していた。よって骨髄-血液-各組織の結合組織へと分配されていることが推察された。マトリックスとの結合様式も特徴があり、線維芽細胞のように紡錘形を保って接着しておらず、細胞は球形を維持しごく限局した部位で結合していることも分かった。ただ血液由来のMuse細胞は培養において接着性が低い特徴が見られた。個体を形成する組織だけでなく、臍帯組織にもMuse細胞が確認され、多能性クラスターの形成や3胚葉分化が確認された。組織恒常性においてはマウスの慢性腎不全モデルにおいて静脈投与したヒトMuse細胞が傷害糸球体に生着し、自発的に足細胞、メサンギウム細胞、血管内皮に分化して機能回復をもたらすことが分かった(JASN,2017)。劇症肝炎(Cell Transpl,2016)、脳梗塞(Stroke,2017; Stem Cells,2016)、心筋分化(Cell Transpl,in press)に関しても成果が発表された。
