Outline of Annual Research Achievements

血液脳関門、髄液脳関門を透過し中枢神経系の広範な領域の神経細胞に目的の遺伝子を送達可能な改良型アデノ随伴ウイルス（AAV）ベクターを応用して、認知症、筋萎縮性側索硬化症（ALS）、脊髄小脳失調症の病態の解明と新規遺伝子治療法を開発することを目標に研究を実施した。シカゴ大学との共同研究により、脊髄小脳失調症6型（SCA6）に対してmiRNAを発現するAAVベクターを使用した遺伝子治療を開発した。SCA6は、日本では優性遺伝のSCAのうち2番目に多く、転写因子α1ACTの遺伝子の塩基配列の一部が異常に伸長している。伸長した配列を伴うα1ACTを搭載したAAVベクターをマウスの小脳に注入し、運動失調を示すSCA6モデルマウスを作製した。このマウスにα1ACTのmRNAから蛋白質への翻訳を選択的に抑制するmiR-3191-5pを搭載したAAVベクターを同時に導入すると神経細胞の脱落が抑制され運動機能が改善した。東京医科歯科大学との共同研究により脊髄小脳失調症１型（SCA１）に対して、DNA修復分子のRpA1を搭載したAAVベクターによる遺伝子治療を開発した。SCA1の原因遺伝子であるAtaxin-1のknock-inマウスモデルのくも膜下腔にRpA1を発現するAAVベクターを投与すると、Purkinje細胞のspineの形態変化が回復し運動機能が改善した。2000年にAAVベクターにより酵素遺伝子を脳内に導入したカニクイサルの組織解析を行い, 炎症反応などを伴わず挿入した遺伝子が神経細胞で15年以上経ても発現していることを確認した。
