Effects of zinc ion on the cytotoxicity of chemicals

Outline of Annual Research Achievements

多くの化学物質の細胞毒性に酸化ストレスが関係している。それらの細胞死に於ける亜鉛の毒性科学的役割を明らかにした。活性酸素種(本研究ではH2O2)の酸化ストレスによる細胞死に細胞内Zn2+は促進的作用という実験成績を得て、さらに細胞死メカニズム解析を進めた。そして、上記のストレスによる細胞膜Zn2+透過亢進および細胞内Zn2+遊離促進のメカニズムを明らかにした。また、温度依存性Zn2+透過機構が細胞膜に存在すること、細胞内チオール量と細胞内Zn2+濃度変化に逆相関があることを明らかにした。上記のデータを基盤にして、各種の化学物質の亜鉛イオン依存性細胞毒性について解析を行い、以下の実験成績を得た。生物殺戮な化学物質(殺菌剤、防腐剤、農薬など)については細胞内亜鉛イオン濃度を変化を伴う細胞毒性が広く認められた。よって、カルシウムイオンと同様に亜鉛イオンも細胞死メカニズムに深く関与していることが示唆できた。上記の生物殺戮な化学物質以外、コーヒー飲料成分や細菌のクロラムセンシング分子にも細胞内亜鉛イオン濃度上昇を起こさせる作用が確認できた。但し、ヒドロキシヒドロキノンでは亜鉛イオン依存性・非依存性の両方の細胞毒性が考えられた。通常、化学物質が誘発する酸化ストレスによる細胞死は細胞内亜鉛イオン濃度上昇により促進する。しかし、細胞内亜鉛イオン濃度上昇や細胞内チオール低下を起こす農薬ジラムが過酸化水素による細胞死を抑制するという現象が確認できた。即ち、細胞死に関係するような要因については促進的な現象が認められているにも関わらず、細胞死が抑制(もしくは遅延)されるという擬似的な細胞保護作用が起こってしまった細胞毒性という分野では見逃されてきた現象と考えられる。
