Outline of Annual Research Achievements

【研究目的】多くの動物は、種の保存や生命の危機回避において、嗅覚を通じて外界から様々な情報を得ている。フェロモンや匂い物質といった化学物質がその情報の担い手であり、情動や行動、生理的変化を引き起こす。研究代表者らは、連携研究者との共同研究のもと、メスマウスの性行動を誘発するペプチド性フェロモンESP1について、物質同定、受容体、性行動に至る一連の構造機能解析を展開してきた。本研究は、これらの知見を発展させ、性行動を誘引するフェロモンや匂いを識別する神経伝達経路をMRIによって包括的に明らかにし、可視化することを目的とする。

【研究実績】ジャコウジカのオスから分泌され、メスを誘引する匂い物質について、マウス脳内における匂い刺激応答を、機能的MRI手法の１つであるBOLD法により捉えることに成功した。１分間隔の繰り返し匂い刺激に呼応して周期的に信号変化が生じる脳内部位を抽出する解析手法を確立し、これを適用することにより、応答部位を同定した。嗅覚伝導路である梨状皮質に加えて、脳深部の領域である外側視床下部や扁桃体にも応答が見られた。また、通常、MRI法において動物は麻酔下において撮像するが、麻酔下動物では捉えられない嗅覚刺激応答を観測するため、覚醒下の動物においてもMRI撮像できる手法の技術開発を進めた。
