狂犬病発病犬の皮膚組織を用いた安価で迅速な確定診断法の開発

Outline of Annual Research Achievements

昨年度は、フィリピンの熱帯医学研究所（RITM)の協力を得て狂犬病発病犬の材料採取を行った。７６頭のサンプリングが終了した。これまで狂犬病発病犬9頭の顔面皮膚を用いたウイルス抗原の局在と今後の死後診断的価値について検証しているが、今後数を増やして検索を継続していく予定である。これまでの結果、9頭全ての洞毛の環状洞の深さに位置する外根鞘の基底層に明瞭なウイルス抗原陽性像が観察されている。同領域の細胞は、抗サイトケラチン20及びウイルス抗体（anti-P抗体）に共陽性を示し、この細胞群がメルケル細胞に該当するものと判断された。本研究によって、狂犬病ウイルスが脳に到達した後に知覚神経を下降し、口唇皮膚に到達し、洞毛のメルケル細胞群において多数増殖すること、並びに洞毛が脳組織に代わる死後の組織診断の材料として極めて有効であることが推測された。本研究内容は狂犬病流行国（発展途上国）に安価で簡便且つ安全な死後診断法を提供するばかりではなく、狂犬病発病犬の試料採取者並びに実験従事者への感染リスクの軽減に大きく寄与するものと思われる。従って、本年度は検体数を増やし、発病犬の洞毛の死後診断的価値について精査したい。なお、昨年度の研究成績は第156回日本獣医学会学術集会（2014.9.16-19, 北大）および2015年1月に開催された17th International Conference on Emerging Infectious Diseases(2015.1.26-29, 台湾)においても発表している。

Reason

2015年1月に病理専攻生2名（博士課程1名、学部生1名）とフィリピンに渡航し、狂犬病発病犬のサンプリングを行った。脳と唾液腺の一部を用いて蛍光抗体法で診断した上で、10％緩衝ホルマリンに固定し病原性を完全に失活させてから病理検索用のサンプリングを行った。
発病犬の捕獲と回収のため、現地関係者とRITMのスタッフの協力を得る必要があったが、皆様から献身的な支えを頂いて計画通りの研究を進めることが出来た。

Strategy for Future Research Activity

病理学的検索：10％緩衝ホルマリン固定のCNS(大脳、海馬、小脳、脳幹、下垂体)、頭部抹消組織（下顎腺、耳下腺、鼻腔、舌、耳、眼球、歯、頭蓋骨）および皮膚組織を用いて切り出しを行った後、組織片をパラフィン包埋する。定法に従いパラフィン切片を作製し、ヘマトキシリン・エオジン染色(HE染色)を施し、病理組織学的検索を行う。

ウイルス抗原および免疫細胞の特定：皮膚組織、脳および唾液腺におけるウイルス抗原の局在について各種特異抗体を用いて免疫組織化学的手法で確認する予定である。

旅費と消耗品の購入費は他の研究費に充てたためである。

昨年度は機材購入が終了したので、今年度は消耗品、国内外の旅費などに当てたい。
