分裂酵母モデル生物を用いた細胞内輸送により調節されるタンパク質リン酸化酵素の探索

Outline of Annual Research Achievements

本研究は分裂酵母モデル生物を用いて、細胞内シグナル伝達経路と細胞内輸送システムの関係を明らかにすることを目的としている。
昨年度に引き続き、分裂酵母キナーゼノックアウトコレクションを用いて、カルシニューリンとPmk1 MAPキナーゼ経路の拮抗的な関係を利用した独自の遺伝学的アプローチを駆使し、表現型を指標としたスクリーニングにより得られた結果をもとに解析を進めた。昨年度、オートファジーに関わるキナーゼAtg1とPmk1 MAPキナーゼとの関わりについて、興味深いデータが得られたことより、Atg1以外のオートファジーに関わる因子についても着目した。オートファジーに関わる因子とPmk1 MAPキナーゼとの関わりについて、オートファジー関連因子破壊株の表現型、遺伝学的アプローチ、細胞内カルシウム濃度の観点から解析し、Pmk1 MAPK構成因子破壊株と同じ表現型を示すオートファジー関連因子破壊株を取得するなど興味深い知見を得た。
また、Pmk1 MAPキナーゼシグナルを負に制御するSkb5を同定し、MAPキナーゼキナーゼキナーゼであるMkh1の細胞内局在を制御することを見出した。タンパク質リン酸化酵素であるMkh1の細胞内局在を制御する因子の発見は、細胞内シグナル伝達と細胞内輸送システムの関わりを明らかにする上での有用な情報となり得る。
