Outline of Annual Research Achievements

上皮細胞は、形態および機能的に極性の分化を示す。すなわち、上皮細胞においては隣接する細胞とのJCを境に細胞膜がapical 面とbasolateral 面に分けられ、細胞質内小器官も不均一な分布を示す。このように正常上皮細胞では、生理的機能を果たすための極性（非対称性）が生じており、隣接する細胞とともに秩序だった組織を形成している。細胞における極性の形成と維持は，生物のボディプランニングや細胞の正しい機能のために不可欠である．atypical Protein Kinase Cλ/ι（以下, aPKCλ/ι）はPAR3, PAR6などの蛋白と結合し細胞極性の形成，維持に重要な役割を果たしている（aPKC/PAR系）。これに対して癌組織では、秩序だった極性が失われ、不規則な組織構築が見られる。病理学では癌の組織診断に際し，構造異型、細胞異型を示標としてきたが、極性の異常は前者を反映している．
本研究で、われわれは免疫組織化学染色を用いて、膵癌、および子宮頸がん前癌病変であるcervical intraepithelail neoplasia (CIN)について検討した。膵臓癌の発生、進行過程でaPKCλ/ιが高発現すること、CINのグレードが高くなるほどaPKCλ/ιが発現が増強、核内に移行することを示した。これによって癌の発生進展にaPKCλ/ιが関与していることが知られた。今後は診断マーカーとしてのみならず、治療標的としての利用法を検討していく。すなわちaPKCλ/ι阻害剤による制癌効果を検討する。また他のがんにも検討を敷衍する。
