Outline of Final Research Achievements |
In the present research, we found for the first time that scaffold protein, ubiquilin2 is mainly involved in this reaction. In vitro experiments using UC lines clearly showed the gene silencing of ubiquilin2 destabilized GRP 78 protein expression, whereas increased upstream molecule of MAP kinase, ASK-1, which could strengthen MAP kinase activation by endogenous ROS and apoptosis induction in carcinoma cells. Such a role of ubiquilin2 in UC survival was proved by nude mice model with implantation of UC in the urinary bladder and in vivo treatment of ubiquilin2 siRNA technique. In addition, immunocytochemical labeling of ubiquilin2 improved both sensitivity and specificity of urine cytology.
