Outline of Annual Research Achievements

本研究は、新規血管作動性ペプチド８候補（Cardiotrophin-1、Fetuin-A、Kisspeptin-10、Stanniocalcin [STC]-1、STC-2、Tumor Necrosis Factor-Stimulated Gene-6 [TSG-6]、Omentin-1、Urocortin-1）の動脈硬化制御作用を世界で初めて明らかにした。動脈硬化に対しCardiotrophin-1、Fetuin-A、Kisspeptin-10、STC-2分画は促進作用があり、STC-1、TSG-6、Omentin-1、Urocortin-1は抑制作用があった。剖検症例の冠動脈硬化病変における上記ペプチドの発現量を検討したところ、Kisspeptin-10をはじめ悪玉ペプチドは病変部に強発現しており冠動脈疾患(CAD)のリスクファクターであることが証明された。
本年度においては、上記候補ペプチドのCADを反映するバイオマーカーとしての可能性を検討するため、急性冠症候群患者135例、非CAD患者24例、健常者28例における血中濃度を測定した。急性冠症候群発症時の血中濃度は、悪玉のKisspeptin-10は増加傾向にあったが、善玉のTSG-6とOmentin-1は予想に反し有意に増加していた。後者２つは動脈硬化を抑制するために反応性に増加していたものと推測された。更に、３年間追跡調査を行って冠動脈イベントや予後との関連を詳細に検討するつもりであるが、CADのバイオマーカーとしてTSG-6とOmentin-1が有力であるが判明した。
ヒト血管細胞（内皮細胞、平滑筋細胞、マクロファージ）及びApoE欠損マウスを用いた実験から、動脈硬化の予防・治療薬の候補としてTSG-6、Omentin-1、Urocortin-1が現在有力であるため、創薬に向けて今後も検討していくことになっている。
