| Outline of Annual Research Achievements |
研究初年度に樹立したアレルゲン特異的ヒトT細胞クローンおよび培養ヒト気管支平滑筋細胞を封入したコラーゲンゲル収縮アッセイシステムを用いて、T細胞培養上清の平滑筋収縮活性の有無を検討した。固相化抗CD3抗体によるT細胞の活性化に依存して、収縮仮性が誘導されることを見出した。抗ロイコトリエン薬、抗ヒスタミン薬、抗コリン薬によって収縮は抑制されなかった。今年度には加えて、幼若マウス気管支平滑筋初代培養細胞を封入したコラーゲンゲル収縮アッセイシステムを樹立した。メサコリン、ロイコトリエンによる用量依存的な収縮を確認した。この系により、27年度に樹立したマウスT細胞クローン移入による抗原特異的遅発型喘息反応モデルに用いたT細胞クローン培養上清の平滑筋収縮活性を評価した。ヒトT細胞クローンと同様に、固相化化抗CD3抗体による活性化依存性に平滑筋収縮活性が誘導されることを見出した。抗ヒスタミン薬、抗ロイコトリエン薬、抗コリン薬によって平滑筋封入ゲルの収縮は阻害されなかった。in vivoアッセイ系であるマウスT細胞クローン移入モデルにおいても、抗ロイコトリエン薬、抗コリン薬の前投与を評価した。約6時間後より、有意な気道抵抗の上昇を認めたが、抗ロイコトリエン薬、抗コリン薬のいずれによっても抑制されなかった。最後に、薬理学的な解析を進める目的に、Magnus管に牽引したモルモット気管リング収縮反応系の樹立を試みた。ロイコトリエン、メサコリン、ヒスタミンによる収縮は検出可能なレベルであったが、良好な感度、再現性が得られなかったため、培養上清の収縮活性検出には実験系のさらなる改良が必要と考えられた。
