| Outline of Annual Research Achievements |
本研究の目的は、周産期新生児感染症において次世代シークエンサーを用いたメタゲノミック診断法を確立し、今まで同定が困難だった病原微生物の同定を行うことを目指した。
平成26年度に、本研究課題内容に関して神戸大学大学院医学研究科遺伝子解析研究倫理審査委員会の承認を得（no. 104、平成26年9月29日付）、研究のセットアップと、すでに病原微生物を同定できている既存サンプルを用いて、次世代シークエンサーによるメタゲノミック解析（後ろ向き解析）で同定できることを確認した。
平成27年度は、正常分娩であっても、分娩方法の違い（経腟分娩か帝王切開か）によって細菌叢が異なるかを検出するために、患者の同意のもと、種々の分娩方法での胎盤および臍帯を計8検体ずつ収集し解析を試みた。また、先天性細菌感染症発症の胎盤を2検体収集し解析を試みた。さらなる次世代シークエンサーでの解析技術の習得を行った。
平成28年度は、次世代シークエンサー解析技術を含む感染症の病原微生物のゲノム解析を行い、周産期新生児および小児感染症に関連する研究成果発表を行った。
今回の科研費での研究を基に周産期新生児および小児感染症領域の病原微生物を検出するための次世代シークエンサーを用いたメタゲノミック診断法の応用や実用化を目指す。また、藤田保健衛生大学総合医科学研究所分子遺伝学教室倉橋浩樹教授や兵庫県衛生研究所感染症部との共同研究体制も構築できた。
