| Outline of Annual Research Achievements |
アトピー性皮膚炎や尋常性乾癬などの皮膚疾患の罹患率は増加傾向にあり、社会問題となりつつある。皮膚は免疫組織としては最大の臓器の一つであるため、皮膚の慢性炎症により局所より産生されるサイトカインが生体の免疫に与える影響は甚大であると思われるが、炎症性サイトカイン過剰モデルが致死性のため、長期間にわたる高炎症性サイトカイン血症が内臓臓器に与える障害を調べた報告は無かった。本研究に於いては、2種類の自然発症皮膚炎モデルを利用し、皮膚炎病変部から産生されるIL-1により高IL-1血症がもたらされ、結果全身の炎症、特に心血管病変・脂質代謝異常・全身性アミロイドーシスを生じるに至る可能性を詳細に検討した。また抗IL-1抗体や炎症性サイトカイン阻害剤の投与により、これらの内臓病変の回避の可能性を検索した。
急性型・慢性型自然発症皮膚炎モデルの組織学的検査、サイトカインの網羅的検索、マウスのCT撮影にての体脂肪率の解析を行った。目標とするサイトカインはIL-1であるが、TNF-αやIFN-γ等も臓器病変に関与する可能性があるため、各々のサイトカイン抗体を長期間投与しての臓器病変の改善効果をみた。乾癬やアトピー性皮膚炎が単に皮膚のみに影響を与えるだけでなく、全身の臓器にも深く関与することが証明でき、Inflammatory skin marchの概念を提唱できた。

| Strategy for Future Research Activity |
マウスのデータの人への応用、また治療効果の人との比較検討を行う。
人に於いて全身炎症を伴った際の合併症、予後の検討も施行よていである。
