Effect of skin-derived antimicrobial peptide cathelicidin LL-37 on the regulation of skin barrier function

Outline of Annual Research Achievements

皮膚は様々な病原微生物から体を守るため、カテリシディンLL-37、β-ディフェンシン(hBD)やS100A7などの抗菌物質を産生するとともに、体内からの水分の蒸散を防ぐことによって、生体を維持するために重要なバリア機能を示す。これら抗菌物質は、抗菌作用の他に、様々な免疫調節作用も有している。アトピー性皮膚炎(AD)と乾癬の両方に角層バリアの欠如があるにも関わらず、抗菌物質は乾癬の皮膚での産生が亢進しており、ADでは減少していることが明らかにされている。ADでは第二種皮膚バリア(タイトジャンクションTJ)の機能に障害があるので、抗菌物質がケラチノサイトにおいて、TJバリアの機能を調整しているのではないかと考えた。
H26年度は、LL-37、hBD-3とS100A7が非定型的プロテインキナーゼC、Rac1、グリコーゲン合成キナーゼ3とMAPキナーゼ経路を介して、ケラチノサイトのTJバリア機能を調節することを報告した。H27年度は、LL-37の特異的受容体を調べたが、同定できなかった。また、LL-37のマウスホモログであるCRAMPのTJバリア機能の調節に対する効果を見られなかった。LL-37のADの痒みに及ぼす影響について調べたところ、LL-37がGタンパク、P2X7受容体とMAPキナーゼ経路を介して、ケラチノサイトにおける神経反発因子であるセマフォリン3Aの発現と産生を増強した。H28年度は、新規の抗菌物質であるAG-30/5CのTJバリア機能に及ぼす影響を見られなかったが、この抗菌物質がMrgX3とMrgX4受容体を介してヒトケラチノサイトやマスト細胞を活性し、創傷治癒を促進することが分かった。
以上より、皮膚が産生するLL-37などの抗菌物質が抗菌作用を持つ他に、ADなどの皮膚疾患におけるバリア破壊、炎症と痒みを抑える可能性があると考えられる。
