Outline of Annual Research Achievements |
To confirm the hypothesis of the involvement of the endocannabinoid system in the favorable recovery profile of propofol, the ability of cannabinoid antagonists to deteriorate its quality has to be examined in several behavioral assays of emotional and cognitive functions. These assays are based upon the recording and subsequent evaluation of animal activity, and any interference with animal activity by anticannabinoid drugs will preclude the correct interpretation of the behavioral results. Therefore, in the first part of this study, we employed the open-field test 1) to determine the optimal experimental time schedule that would allow the evaluation of multiple doses of cannabinoid antagonist in the same animal; and 2) to determine the dose range of cannabinoid antagonist that would have no effect on the total distance travelled by mice in the open-field test. We planned to use these data in the second part of this study to confirm the involvement of endocannabinoid system in the positive affective state produced by propofol in the place preference test. However, this could not be done as planned due to the failure of the dealer company to timely provide a properly made and ready-to-use piece of equipment (CPP box) necessary for the conduction of the above test. After having to participate in the designing and testing of the trial version of the CPP box, we are currently (as of April, 2016) waiting for the delivery of its properly made version, which will also have to be tested before transitioning to the initially planned experimental schedule.

Reason
The research plan for the second experimental year could not be realized due to the failure of the dealer company to timely provide a properly made and ready-to-use piece of equipment (CPP box) necessary for the conduction of the place preference test. After having to participate in the designing and testing of the trial version of the CPP box, we are currently (as of April, 2016) waiting for the delivery of its properly made version, which will also have to be tested before transitioning to the initially planned experimental schedule.

Strategy for Future Research Activity |
In the following year, after the CPP box is delivered, we will have to confirm that this piece of equipment meets our requirements and can be used as it is without further improvements. After this is done, we will be able to start place preference experiments to investigate the involvement of the endocannabinoid system in the positive affective state produced by propofol. Since we are running out of time allocated for the current project, it will be probably necessary to apply for the prolongation of the research period at the end of the upcoming year.

Causes of Carryover |
The amount for the fiscal year concerned has not been completely used due to reasons beyond our control. The remaining amount will be used in the third year of this research project.

Expenditure Plan for Carryover Budget |
This amount will be used for the purchase of supplementary pieces of equipment (lighting lamp for the CPP box) and materials (animals and reagents) necessary for the study.
