Outline of Annual Research Achievements

我々がconditional gene targetingにて確立した、前立腺特異的PTENノックアウトマウスと同じ手法である、Cre-LoxPシステムを用い、PTEN+/+/Trp53f/fマウスとPSA-Cre+マウスを交配することにより、PSA-Cre+/Trp53f/fマウスを作製し、♂PSA-Cre+/PTEN homo deletionマウスと交配させ、PSA-Cre+/PTENf/+/Trp53f/+ (PTEN、Trp53ともにヘテロ欠失型）を確立し、ヘテロ型同士の交配により、PSA-Cre+/PTENf/f/Trp53f/+(PTEN homo p53 hetero deletion)、PSA-Cre+/PTENf/+/Trp53f/f(PTEN hetero p53 homo deletion）、PSA-Cre+/PTENf/f/Trp53f/f（PTEN,p53 homo deletion)の3表現型Colonyを確立した。そのうちPTEN/p53ホモ欠失マウスについて病理組織学的な経時的変化や増殖・進行についてCharacterizeした。このモデルでは、早期から全身転移をきたし、生後60週までに癌死する転移として生存期間をエンドポイントとした研究が可能な実臨床に則したモデルとして有効であることが確認された。このマウスモデルを用いてランダム化前向き試験を模倣した治療研究が可能となり、Preclinical Researchとして新規の治療法の有用性の検討が可能となった。
