Outline of Annual Research Achievements

線維柱帯細胞の貪食能に対する各種薬剤の影響について検討することを目的に実験を行った。ブタの眼から線維柱帯細胞を単離、培養し、継代数4-6回で実験に用いた。また、ヒト由来の線維柱帯細胞を不死化したTM-3も実験に用いた。ブタ線維柱帯細胞の培地を一晩無血清とし、その後蛍光マイクロビーズ(staphylococcus aureus bioparticle)を培地に加え、1~2時間培養した。その後wash outして細胞外のビーズを抗体標識し、細胞を4%パラフォルムアルデヒドで固定した。蛍光顕微鏡で写真撮影し、1細胞あたりの取り込数をオートカウンターにてカウントした。刺激薬剤は、ステロイド薬であるデキサメサゾン、Rho-associated kinase(ROCK)阻害薬であるY-27632、monocyte chemoattractant protein-1(MCP-1)、transforming growth factor(TGF)-β2を用いた。10、100、1000nMのデキサメサゾンで刺激した結果、100nMデキサメサゾン刺激によってコントロールと比較して貪食が低下する傾向にあった。一方で、2.5、10、25μMのY-27632で処理したところ、25mMの濃度で貪食が有意に増加した(P<0.05)。一方で、MCP-1、TGF-β2でも刺激を行なったが、有意な変化はみとめられなかった。ヒト線維柱帯細胞由来のTM-3を用いた同様の実験では、100nMデキサメサゾンによって貪食が低下する傾向があり、10および25μMのY27632で有意に貪食が増加した。以上のことから、デキサメサゾンは線維柱帯細胞の貪食脳を低下させ、ROCK阻害薬は逆にこれを向上させることが示唆された。
