Outline of Annual Research Achievements

Model organism was decided and it was determined that it produced PAH-type catechols during biotransformation of PAHs. Genes involved in catabolism of PAHs by this organism were determined. RT-PCR methods were established by investigating expression of these genes. Assessment of DNA damage by LC/ESI-MS/MS DNA adductomics was conducted using the model organism and model toxicant hydrogen peroxide.

Reason

Determined model organism. Determined that this organism biotransforms PAHs through a PAH-catechol type pathway by analyzing biotransformation products during exposure to PAHs. Established methods for conducting RT-PCR analyses by investigating genes involved in PAH catabolism.

Strategy for Future Research Activity

Continue DNA damage analyses using hydrogen peroxide model toxicant. Begin investigation of DNA damage by using PAH o-quinones. Continue RT-PCR analyses. Develop methods for detecting ROS by fluorescence in model organism. Continue to develop software for processing DNA adductome data.

Causes of Carryover

Budget from 2014-15 was carried over to 2015-16 because it was sufficient for the experiments conducted in 2014-15.
