Outline of Final Research Achievements

The aim of this study was to develop fundamental research protocols for a novel high-throughput screening method in order to identify any defined RNAi combinations to control cell lineages via novel RNAi-based reprograming strategies. Particularly, to develop such a screening method, I combined whole-genomic shRNA lentivirus library and next-generation sequence technique. I developed and fine-tuned several complexed experimental protocols related to Lentivirus production, Lentivirus infection, cell culture, cell-sorting or cell-picking, and next-generation sequencing. I could establish some of the fundamental research protocols to be utilized in the high-throughput screening to search for defined RNAi combinations to induce lineage reprograming.
