| Outline of Annual Research Achievements |
1，2-ジクロロプロパンは様々な塩化有機化合物製造の中間物質として用いられるとともに洗浄剤としても使用されている。2014年、国際がん研究機関（IARC, International Agency for Research on Cancer)は、1，2-ジクロロプロパンをGroup3（分類不能）から、Group1（ヒトでの発がん性あり）に分類しなおした。これは日本における疫学研究に基づく判断である。一方、日本と米国におけるマウスとラットを用いたバイオアッセイ研究は、一部のがんの増加を示しているが、胆管がんそのものの増加を示していない。また、これまでの研究は1，2-ジクロロプロパンの胆管がん誘導作用のメカニズムを十分に明らかにしていない。本研究は、マウスにおける1，2-ジクロロプロパンの胆管上皮細胞への影響を、細胞増殖と細胞死の観点から明らかにするとともに、それらの影響におけるP450の貢献を明らかにした。マウスC57BL/6Jに1，2-ジクロロプパンを1日8時間、週7日、4週間吸入曝露するとともに、一部のマウスにはP450阻害剤である1-aminobenzotriazoleの腹腔注射を毎日行ったうえで1，2-ジクロロプロパン吸入曝露を行った。1，2-ジクロロプロパンへの曝露は、胆管細胞の増殖、胆管細胞のアポトーシス、肝細胞壊死を増加させたが、P450阻害剤1-aminobenzotriazole投与群ではこれらの影響は抑制された。本研究は、動物の胆管上皮細胞において1，2-ジクロロプロパン曝露が細胞増殖とアポトーシスを誘導することを示した初めての研究であるとともに、P450による代謝活性化が、これらの影響に貢献していることを示した。
