遺伝学とプロテオーム解析を組み合わせたDNA損傷応答の新規制御機構の解明

Outline of Annual Research Achievements

遺伝学的な解析が可能であるニワトリDT40細胞を用いて、DNA損傷修復に極めて重要な因子の一つであるBRCA1の相互作用因子を質量分析機を用いて解析を行った。その結果、BRCA1蛋白質はDNA損傷応答に関わる因子だけでなく転写や染色体分配に関わる多くの因子と相互作用することが明らかになった。その中には、今までBRCA1と相互作用するという報告がない因子も含まれている。研究期間中に新規遺伝子の同定まで至ったという点で、当初の目的は達成されたと考えている。現在、その新奇遺伝子の機能解析を実施している。
また研究期間の後半からゲノム編集技術(TALEN, CRISPR)を用いてヒトBRCA1変異細胞と3FLAGエピトープ配列をBRCA1のN末端領域にノックインした細胞を樹立した。このヒト細胞を用いてプロテオーム解析を実施中であり、今後ニワトリ細胞とヒト細胞を比較することにより多機能分子であるBRCA1の機能の全容を明らかにできると考える。
