Outline of Final Research Achievements |
There have been many attempts to make artificial oxygen carriers based on the modification of isolated hemoglobin (Hb). In these attempts, artificial oxygen carrier realizes improved characteristics, avoiding infection, no blood type, and longer shelf life than red blood cells (RBC). In this work, novel large-sized Hb assembly (aligned megadalton Hb; AM-Hb) was synthesized by intermolecular crosslinking of Hbs with 4-armed polyethylene glycol (PEG) to prevent extravasation. The physicochemical properties as oxygen carrier was evaluated for AM-Hb. The molecular size of AM-Hb depends on the molecular weight of PEG linker. AM-Hb has larger viscosity and larger oxygen affinity than those of RBC, Hb, and Hb vesicles.
