Outline of Annual Research Achievements

高齢社会を迎えた我が国や今後迎えるであろう先進諸国において,服薬ノンコンプライアンスの改善は喫緊の課題となっている.本申請課題は,医薬品の苦味を客観的に数値化するための新規味覚センサの開発を目指すものである.味覚センサは,人が各味受容体を有することを模倣し,基本五味毎に対応する脂質高分子膜電極を用い,各基本味の味強度の数値化を客観的に評価する世界初・日本発の膜電位計測型のケミカルセンサである.本申請課題では,医薬品の苦味をヒトの官能値と一致する新規味覚センサの開発を目指し,実施項目①脂質,可塑剤の分布に関する分析,実施項目②脂質高分子膜表面の脂質,可塑剤の分布度合いと医薬品の吸着量の計測,実施項目③脂質高分子膜の膜電位応答と脂質高分子膜の構造との関連性評価の3項目について実施する.本年度は,実施項目③に関して実施した.また,昨年度開発をおこなった人工甘味料による苦味の抑制効果を苦味センサと甘味センサを用いて評価する新規手法の開発も実施した.脂質高分子膜は,昨年度までに検討をおこなった脂質及び可塑剤の種類・量と疎水性度,脂質膜表面電荷密度に関する知見を基に最適化した結果,センサの感度,選択性のみならず耐久性を改善することができ,長期使用に耐えうる脂質高分子膜の作製に成功した.また,医薬品苦味のモデル味物質として用いられる塩酸キニーネと苦味抑制剤として用いられる人工甘味料を混合したサンプルを対象とし,苦味センサと人工甘味料用センサを用い,人の官能値と相関が高い統計モデル式の作成に成功した.
