製薬における品質・生産性向上にむけた多目的プロセス設計手法の開発

Outline of Annual Research Achievements

本研究の目的は、製薬プロセスの品質・生産性向上にむけた多目的設計手法の開発である。H27年度の成果は以下の3点にまとめられる。
(1)注射剤製造のプロセス改善手法の開発:注射剤製造における製品ロスの低減や品質劣化・検査コストを考慮した原料管理、製造リードタイムの改善に関してプロセスを改善するための手法を開発した。このうち製品ロスの低減手法については、H26年度までの研究を発展させ、プロセスパラメータと製品ロスの相関を見ていくことで、ロスの要因に踏み込んでプロセス改善を行えるようにした。手法はバイオ医薬品注射剤を製造する実際のプラントに適用され、プロセス改善を実施した結果、不良品を4分の1程度まで削減することに成功した。

(2)製造プロセスの技術選択支援手法の開発:注射剤製造を対象に、バッチ生産毎に洗浄・滅菌をする従来の「マルチユース技術」と、バッチ生産毎に使い捨てられる新規の「シングルユース技術」を比較評価する手法を開発した。H26年度までは経済性や環境影響に関する評価に焦点を当てていたが、H27年度は製品品質や供給に関わるリスクも今日慮対象に入れている。また、H27年度からは固形製剤に関する技術選択にも着手し、従来のバッチ式技術と新規の連続製造技術を比較評価する手法の開発を始めた。

(3)環境・健康・安全(Environment, Health & Safety: EHS)リスク低減支援手法の開発:低分子原薬製造プロセスを対象に、使用溶媒に関わるEHSリスクを低減するための手法を開発した。H26年度までに開発してきた手法では反応・抽出・晶析・乾燥などの製造主要部分に焦点を当てていたが、H27年度は溶媒の回収や貯蔵、分離といった周辺プロセスも含む形で手法を発展させている。本手法ではプロセス改善を行う際に、製薬産業特有の品質基準を考慮した多目的評価を取り込んでいる。

Reason

研究はおおむね順調に進展している。交付申請書には「プロセスの品質・生産性評価指標の開発」、「製薬固有の条件を考慮した解析手法の構築」、「課題発見・解決の業務モデル構築」の3点を具体目的として示した。1点目に掲げた評価指標開発については、製品ロスの優先順位や技術選択評価、EHSリスク評価について、それぞれ新規な指標を開発した。2点目の解析手法としては、製造工程の数学的記述方法や、品質基準を制約条件として取り込んだ多目的評価手法を構築した。これらの結果をもとに、最終年度であるH28年度に3点目の業務モデル構築に着手しようとしている。テーマについても、当初注力していた注射剤製造から原薬製造、さらには固形製剤製造に展開しており、製薬プロセスの主要部を対象として含む研究になってきている。

Strategy for Future Research Activity

今後の推進方策は次の4点にまとめられる。(1)プロセス改善手法の発展:バッチごとの製造データを詳細に分析し、改善の対象をプロセスの運転と設計に分割していくことで、より本質的なプロセスの改善を可能にしていく。(2)技術選択支援手法の発展:注射剤製造のシングルユース・マルチユース、固形製剤製造の連続・バッチについて、評価項目を拡張し、実用性を高める。(3)EHS評価手法の発展:手法の対象を、製造の主要部から周辺部、さらには複数プロセスを内包するサイト全体に拡張していくことで、より実態に即した評価に基づくプロセス改善を可能にする。(4)設計手法への展開:これまで行ってきたケーススタディの知見をもとに、運転中のプロセスでの課題がそもそも起きないようなプロセス設計のあるべき姿を探る。
