Research proposal for this budget year planned analysis of luminescence genes expression in V. chagasii mutants. However, creation of the mutants, planned for H26 budget year, was not yet completed. Therefore, we continued creation of V. chagasii mutants.
In the H27 budget year we continued analysis of QS regulatory genes of two strains of V. chagasi, 21N-12 and SB-52. We cloned luxO and luxR genes of V. chagasii for use in creation of deletion mutants. We also established and refined a method for creation of V. chagasii mutants. The procedure involves introduction of suicidal plasmids into V. chagasii. This is the first time a method for creation of V. chagasii mutants was developed.
