Analysis on the mechanism of immune regulation by CD4+ conventional dendritic cells in the development of autoimmune diseases

Outline of Annual Research Achievements

平成28年度はDCIR2欠損マウスにおける自己免疫病態の解析を行った。具体的には野生型（WT）マウス、DCIR2欠損マウスに自己免疫性脳脊髄炎（EAE）を発症させ、病態評価および自己抗原特異的T細胞活性化能について比較検討を行った。病態評価では、神経性自己抗原（ミエリンオリゴ糖タンパク質ペプチド; MOGp）の免疫後より経時的にEAE発症率および臨床スコアを測定した。その結果、WTマウスと比較して、DCIR2欠損マウスではEAE病態の増悪化が認められた。また、自己抗原特異的T細胞活性化能の評価では、免疫14日後に各マウスからCD4陽性T細胞を精製し、MOGpを抗原としたWTマウス由来脾臓樹状細胞との共培養を行った。培養後、CD4陽性T細胞活性化能の指標としてMOGp特異的T細胞増殖反応（[3H]-TdR取り込み能）を測定した。さらに、その培養上精中のIFN-gとIL-17A産生量をEILSA法により測定した。WTマウスのCD4陽性T細胞と比較して、DCIR2欠損マウスのCD4陽性T細胞ではMOGp特異的T細胞増殖反応およびIFN-g、IL-17A産生量の亢進が認められた。以上の結果から、CD4陽性通常型樹状細胞はDCIR2の制御下において自己抗原特異的T細胞応答を抑制し、自己免疫病態を制御することが明らかとなった。
