Outline of Final Research Achievements |
It is very meaningful in the field of orthopedic surgery to aim for early regeneration induction by drug therapy against peripheral neuropathy requiring a long treatment period. GPR30 is thought to be involved in the signaling of the myelination mechanism as a membrane type receptor. Establish effective primary culture system of dorsal root ganglia and spinal cord progenitor cells. For the neurons, dorsal root ganglia (DRG) was selected and extracted from fetal mice. Furthermore, for the primary culture of the spinal cord anterior horn cellular motor neuron, it was extracted from the rat spinal cord using a special kit (NycoPrepTM). To extract Schwann cells, rat sciatic nerves were collected, subjected to dissolution treatment, and extracted. Separation of cell bodies and axons AXIS and Campenot chamber were used to establish a culture system. As an observation method, the timing of observation with a confocal laser microscope and the method of quantification were adjusted.
