網膜色素上皮細胞における上皮間葉転換と生物学的意義

Outline of Annual Research Achievements

網膜色素上皮細胞の上皮間葉転換 (EMT) は増殖硝子体網膜症などの線維性病変の原因の一つであり、我々はTNF-αが誘導するEMTメカニズムに注目している。このTNF-αが誘導するEMTにおいて、FERMドメインを持つMerlinが非刺激下ではCD44と会合して細胞膜に局在する一方で、持続的なTNF-α刺激により低下することも見出した。Merlinは非刺激下のRPEではCD44と会合し、TNF-α刺激下で会合が減少していた。Merlin siRNAの結果、ERMのリン酸化が亢進し、CD44とリン酸化ERMは共局在し、ヒアルロン酸結合能が亢進した。さらに、Merlin siRNAはTAK1ーp38MAPKシグナル活性化に関与を誘導し、細胞増殖とEMT誘導に必須であった。これらの結果はRPEの上皮間葉転換誘導に重要な経路を特定するものであり、EMTが関連する線維性疾患の新たな標的となる可能性がる。

Reason

TNF-αが誘導するEMTにおいてMerlinの役割をin vitroにて明らかにし、その成果はInvestigative Ophthalmology & Visual Scienceに2015年3月受理された。また、硝子体サンプルとEMTシグナルの活性化について解析を行った結果は2016年1月にBiochmical and Biophysical Research Communicatonsに受理された。

Strategy for Future Research Activity

Merlinの下流で活性化を認めたTAK1に注目し、その阻害薬の効果をin vivo増殖硝子体網膜症モデルでも検討する。
