Research Abstract |
In order to clarify the mechanism of peptidergic bio-communication system involved in the neuronal and hormonal network, we have performed a systematic survey for so far unidentified bioactive peptides in the mammalian tissues. By using a smooth-muscle stimulant or relaxant activity as an tentative index for bioactivity, neuromedin K, L, B, B-30, B-32, C, N, U-8 and U-25 have been identified in porcine brain and spinal cord. Their specialized localization in the central nervous system and peripheral tissues suggested their physiological importance in the bio-communication system. By the similar bioassay for relaxant effect on chick rectum preparation, human and rat atrial natriuretic polypeptides (ANP) have been purified from atrial tissues and sequenced. Identification of ANP demonstrated that the mammalian heart functions not only as a pump impelling the blood, but also as an endocrine organ that secretes the hormone controlling body fluid volume, electrolyte balance and blood pressure. Cloning and sequencing of cDNA encoding ANP precursors in mammals has also been achieved. In ovarian follicular fluid of pig and beef, inhibins of 32K daltons have been purified to homogeneity. Identification of inhibins reveals that inhibin regulates the FSH release from pituitary. On the other hand, processing enzymes, which generate the mature bioactive peptides from their precursors, have a critical function to control the synthesis and secretion of peptide hormones or neuropeptides. The proteolytic enzyme specifically cleaving the dibasic processing signal has been first purified from yeast. Purification of C-terminal amidating enzyme from frog skin is now going on.
