Research Abstract |
The purpose is to investigate glycoconjugates of the nephron in an experimental pyelonephritis and in human pyelonephritis. Several kinds of peroxdase labeled lectins were used to identify lectin binding sites in the nephrons. Experimental pyelonephritis in rabbits induced by E.coli were employed for the study. Animals were sacrificed 1,2,3 days, 1,2,4,8,12,24 weeks and 2 years after the inoculation. Normal and ureter stenosed animals were studied as controls. In normal rabbit kidneys, PNA showed strong affinity to thin loop, thick ascending loop of Henle (TcLH) and collecting ducts in the cortex. Although no staining was observed in TcLH in the outer medulla of the normal kidney, positive staining with PNA was obviously reviewed there in acute and chronic pyelonephritis. PNA binding on TcLH was observed 3 hours after complete ureteral obstruction. It is suggested that galactose B(1->3)N-acetyl-galactosamine which specifically binds to PNA appeared in TcLH. The results present the possibility for susceptibility to bacterial infection in TcLH as far as an experimental pyelonephritis in rabbits is concerned. Based on the finding of DBA and PNA staining, thyroidation of tubules in chronic pyelonephritis may consist of TcLH and collecting duct of the outer medulla in rabbit's kidney, although it has been thought that proximal tubules change to thyroidation according to the evolution. Nine cases of nephrectomized human specimens clearly disclosed same findings. P and type 1 fimbriae were investigated in 153 strains isolated from urogenital infections. No significant correlation among fimbrial type, hemoagglutination titer and hemolysin production was noticed. No significant glycoconjugates distribution was observed in the testis following vas ligation. However, flagellar damage in the epididymal duct may cause epididymitis in rabbits.
