Research Abstract |
As the mutagenicity for gaseous or volatile compounds has not been established, we have developed a simple method by bubbling for detecting the mutagenic activities in these compounds. Vinyl chloride and ethylene oxide, for example, were tested by this simple method. Vinyl chloride(VC)(100%) was absorbed into dimethylsulfoxide(DMSO) in a tube by bubbling for 10-15 minutes. Then 0.1ml of the above DMSO, 0.1ml of a fresh cultured tester strain (E.coli WP2uvrA/pKM) and 0.5ml of S9Mix were mixed in a gas tight vial, shaked at 37 C for 30-120 minutes and poured with 2ml of top agar onto a minimal glucose agar plate. Trp revertants were scored after incubation at 37 C for 48 hours. On the other hand, the concentration of VC in the above DMSO was measured by gaschromatography. The mutagenicity test of ethylene oxide(EO)(12%) was performed by this method. EO was absorbed into 10ml of ethylalcohol. The mutagenicity test for EO was carried out by the same method using S.typhimurium TA100 without S9Mix and preincubation. The quantitative analysis of EO was performed by gaschromatography. The positive results with dose-response effects were observed both VC and EO. The regression curves are Y=0.2X + 205.3 in VC and Y=0.157X + 157.3 in EO (p 0.001). This bubbling method is useful and simple for detecting mutagenic activities in gaseous compounds, quantitatively.
