Research Abstract

KLH抑制T細胞抗原レセプタ-α鎖遺伝子の解析を行い次の結果を得た。1.新しいTCR遺伝子ファミリ-(Vα14)を発見した。2.Vα14を遺伝子プロ-ブにしたサザンブロット解析から、ラボマウスは3種類に分類できる。そのうちC57BL系はアジア型野性マウスタイプと、その他のラボマウスはヨ-ロッパ型野性マウスタイプを示した。3.Vα陽性α鎖は正常マウスにおいても90%以上がJα281と結合し、VJ結合はほとんど1塩基挿入のN領域(グリシン)で構成されるhomogenous receptorであり、positive selectionの結果であると考えられた。4.V14J281α鎖は全α鎖の1.5%をも占め、高頻度に発現していた。5.このリガンドはキメラマウスの実験から未知の自己成分でマウス亜種特異的に発現する分子であることが判明した。6.このα鎖の選択は胸腺非依存性にもおこなわれていることが、ヌ-ドマウスや胸腺摘出X線照射骨髄マウスを用いた実験から示唆された。以上のことから、抑制T細胞は自己反応性レパトアに属し、自己寛容の維持に重要な働きを持つと考えられた。
