Research Abstract

Formation of selenotrisulfide has been known as a process of metabolism of selenium. However, characterization of selenotrisulfide formed from selenite and biologically relevant thiols such as glutathione and cystein was not achieved, because of their comparative lability. We found that selenite and penicillamine formed stable selenotrisulfide which could be isolated in pure state. This selenotrisulfide was fully characterized by various spectroscopic methods. In addition, the occurrences of thiol exchange reactions were confirmed in the reactions of selenotrisulfide in the presence of other thiols. These reactions may serve as the model reaction for the metabolic process of selenium. Development of accurate and simple method of analysis which is applicable to environmental and biollogical samples has been of great demand for the enhancement of the development in the chemical studies of the biological activity of selenium. We applied the formation of stable selenotrisulfide to develop a preconcentration and a determination method for the trace amount of selenium. We have established a system of analysis for selenium, consisting of preconcentration by use of anion-exchange resinloaded with bismuthiol-II sulfonate to form stable selenotrisulfide selectively on the resin, and determination by HPLC based on the formation of stable selenotrisulfide of penicillamine, which is detected fluorometrically through a fluorescent derivative. The system of analysis gave satisfactory results in the determination of selenium in some environmental and biological samples with certified values. The applicability of our method of analysis to the environmental and biological samples has been thus confirmed.
