Research Abstract |
It is well established that precipitate of altered hemoglobin known as Heinz bodies in erythrocytes and that the life span of erythrocytes in the circulation is shortened by the presence of these precipitates. However, the formation mechanism of Heinz bodies is not fully anderstod. Oxidation of protoporphyrin of heme was proposed to alter the contact to lead to destabilization of hemoglobin molecule. The molecule diminishes its resistance to oxidation by oxidants produced concomitantly by the reaction of oxygen with hemoglobin in erythrocytes, and leads to oxidative denaturation to form Heinz bodies which associate with the life span of erythrocytes. An oxidation product of protoporphyrin was isolated by treatments with acidicmethanol of intact human erythrocytes. The compound was analyzed by thin-layer chromatography and high-performance liquid chromatography. Results showed to be dimethlester of propentdyopent with methl and propionic acid residues. The propentdyopent was derived by odxidation of heme with hydrogen peroxie in chemical reaction. Prepartion of many standard compounds used for above analysis also was described in this paper.
