Research Abstract	本研究は,ATP分解と収縮力の発生という,生命現象における最大の課題の解明に関わっている. 本研究では,各分担者が以下のいくつかの例に示すような幅広い実験系を用いて,互いに議論を深めつつ研究をすすめた. 1.収縮タンパク質および収縮制御タンパク質を対象とする研究. 柳田(大阪大学)は,螢光で標識した1本のアクチン線維が,ミオシン線維上を運動する様子を顕微鏡下で直接見ることのできる実験系をつくり,これによって1回のATP分解反応がおこるとアクチン線維は最大50ナノメートル滑走することが示された. この滑走は負荷の大きさによって一定ではなく,負荷によって滑走の距離が変る. このことは生理学上古くから知られてきたFennの効果とよばれる筋収縮における基本的性質が,実は分子のはたらきに存在することを示しておりたいへん重要である. 2.細胞膜剥離標本を対象とする研究. 飯野(東京大学)らは,細胞膜剥離標本について,無機リン酸濃度の与える効果を調べた. 無機リン酸はアクトミオシンによるATP分解産物の1つであり,現在の収縮の分子モデルの検証において,無機リン酸濃度が収縮に与える効果を知ることは大変重要である. 結果の確立にはさらに研究を継続させねばならない. 米谷らは同様の実験系を用いてエチレングリコールの効果を調べた. その結果はこの物質は上にのべた分子モデルにおいてパラメーターの1つを修飾していることを示唆しており,モデルの検証にとって重要な研究結果である. 3.単一筋線維および全筋を対象とする研究. 山田他(大分医大),山田と杉(帝京大)および富田ら(名大)はリンNMR法による生きた筋についてのエネルギー変換過程の研究を行ない,いくつかの重要な発見がなされた. 松原ら(東北大)はX線回折法により,同様に生きた筋について収縮により分子的変化を追求しつつある.
