Research Abstract

1.ウシのインヒビンのRIA系を用いてヒトのインヒビンを末梢血中レベルで測定することが可能となった。このRIA系を用い種々の生理的あるいは病態に伴うインヒビンの変動を明らかにすることができた。(イ)正常月経周期中では卵胞期初期では低く、徐々に増加する。このインヒビンの増加に伴いFSHが減少することを証明した。また黄体期にインヒビンが上昇することから黄体期のFSHはインヒビンにより抑制されていることが明らかとなった。(ロ)男性において血中インヒビンの日内変動が明らかにされた。早朝よりインヒビンは上昇し9時にピ-クとなり、その後ゆっくり下降し22時に最低値となった。また、男性の加令に伴う変動を調べた。その結果、インヒビンは20歳台で最高値を示し、そ後徐々に減少し、60才以降に急激に減少した。また、低機能の睾丸を持つ男性のインヒビンレベルは低く、老人のそれより低いことが明らかになった。(ハ)妊娠時においては、インヒビンは徐々に増加し、8〜10週にピ-クを示した。一度減少したインヒビンは30週以降にピ-クを示した。このような二相性のインヒビン分泌から、妊娠中のインヒビンの分泌源が黄体と胎盤の二ケ所にあることを示唆する。分娩後速やかに血中インヒビンは減少した。(ニ)新生児の血中インヒビンは男子において有意に高いことが明らかとなった。
2.アクチビンの卵巣における作用をin vivo、in vitroの系において明らかにした。(イ)アクチビンは顆粒膜細胞中のFSHレセプタ-を誘導し、FSHの作用を増強する作用がある。その結果、LHレセプタ-の発現が促進され、P_4の分泌が増加した。幼若のメスラットにアクチビンを1週間投与すると用量反応的に卵巣の重量が増加した。その卵巣を組織学的に調べるとアクチビンにより卵胞が発育し、成熟卵胞が2倍に増加していた。
