Research Abstract |
SINCE THE ESTABLISHMENT OF THE TWO STAGE CARCINOGENESIS, VARIOUS TUMOR PROMOTERS HAVE BEEN REPORTED. EFFECTS OF TUMOR PROMOTING AGENTS ON THE MALIGNANT TRANSFORMATION OF THE HYDATIDIFORM MOLE CELLS ( BM-34 ) MAINTAINED FOR OVER TWO YEARS IN CULTURE, WERE EXAMINED. IN VITRO COLONY ASSAY SYSTEM IN SOFT AGAR AND XENOGENIC TRANSPLANTATION METHOD WERE USED TO PROVE WHETHER MALIGNANT TRANSFORMATION OF HYDATIDIFORM MOLE CELLS TREATED WITH THE PROMOTERS ( TPA: 12-O-TETRADECANOYL-PHORBOL-13-ACETATE ), ( TLC: TELEOCIDIN ), ( VITAMIN D DERIVATIVES : 1ALPHA,25-(OH)_2 D_3), HAD OCCURRED. THE HYDATIDIFORM MOLE CELLS WERE TREATED WITH TPA (0.1,1.0,10,100 ng/ml), TLC (1.0,10 ng/ml), VITAMIN (0.1,1.0,10 hg/ml) FOR VARIOUS LENGTH OF TIME (8,10,12,16,20 WEEKD). WHEN THE TREATED CELLS WERE SEEDED IN SOFT AGAR UNDER PRESENT TREATMENT CONDITION WITH TPA, TLC AND VITAMIN D , THE PLATING EFFICIENCY OF HYDATIDIFORM MOLE CELLS TREATED WITH TPA AND TLC FOR OVER 12 WEEKS INCREASED SIGNIFICANTLY IN COMPARISON WITH CONTROL, WHEREAS THAT OF THE CELLS TREATED WITH VITAMIN D FOR 12 WEEKS FROPPED IN INVERSE PROPORTION TO EACH CONCENTRATION OF THE DRUG. IN ADDITION, WE DID NOT OBTAIN ANY PROGRESSIVELY GROWING TUMOR EVEN IF WE HAD ATTEMPTED TO TRANSPLANT THESE CELLS INTO NUDE MICE.
