光力学的癌治療における新しいより有効な光感受性物質の利用と開発に関する研究

Research Abstract

1.Alー、Gaー、Inーフタロシアニンのレ-ザ-照射後の光力学的抗腫瘍効果の比較では、Alーフタロシアニンにより最も強い効果が認められた。またAlーフタロシアニンはHpDに比較し皮膚残留性が少なく、かつ選択的腫瘍集積性が高いという結果が得られた。
2.HpDおよびAlーフタロシアニンを用いたマウス皮膚における光過敏反応の検討では、長波長紫外線照射ではHpDの皮膚過敏反応はAlーフタロシアニンより強く、可視光線照射では両者に差は認められなかった。
3.透過型電子顕微鏡を用いたラット正常膀胱壁への光力学的治療の検討では、照射部の粘膜上皮細胞は上皮基底膜から剥離し、滑面小胞体の内腔の拡張、細胞質の膨化が認められた。また粘膜上皮および粘膜固層の毛細血管内皮のミトコンドリアの空胞化、クリスタの溶解および変性が認められた。
4.HpD、Hpオリゴマ-、ゲルマニウム・ヘマトポルフィリン誘導体の光増感剤としての蛍光寿命測定での検討では、Hpオリゴマ-はHpDと同程度の長寿命成分を有していた。Hpオリゴマ-の組織内への取り込みがHpDに比べ良好であることから考えると、Hpオリゴマ-はHpDと同様あるいはそれ以上に光力学的反応性が高いと期待された。
