| Research Abstract |
1.IL-1に関する研究
ネコ電線性腹膜炎(FIP)羅患ネコの腹腔滲出細胞(PEC)の培養上清中には高いIL-1活性が認められることが明らかになり、またそのIL-1活性を含むRECの培養上清は健常ネコの抹消血好中球(PBN)に対してchemoattractontとして作用することが判明した。健常ネコのPBNのPEC培養上清に対する遊走は、Zymosanで活性化した血清に対する遊走に比べ弱い傾向が認められ、各chemoattractontに対する感受性も異なっていた。
2.IL-6に関する研究
FIP羅患ネコの血清および腹水中のIL-6活性をハイブリド-マ細胞の増殖を指標として測定した結果、非常に高い活性が認められ、またPECの培養上清にも同様に高いIL-6活性が検出された。腹水中のIL-6活性はゲル濾過法で分子量30000から40000に相当する分画に検出され、イオン交換クロマトグラフィ-で塩濃度0.2から0.3Mにかけて溶出された。腹水中のIL-6活性は血清アルブミンノグロブリン比と逆相関し、FIP羅患ネコにおける高γ-グロブリン血症や急性相蛋白の増加にIL-6が関与することが示唆された。
3.IL-2レセプタ-(IL-2R)に関する研究
ネコのレトロウイルス感染症における免疫異常をIL-2Rの面から解析するためにネコのIL-2Rの鎖に対するモノクロ-ナル抗体を作製し、さらにネコ白血病ウイルス(Felv)産生リンパ肉腫細胞株におけるIL-2Rの発現を検討した。9F23抗体は^<125>I標識IL-2と結合するリンパ球表面上の分子量52000の蛋白を免疫沈降することからネコのIL-2Rの鎖を認識することが確認された。またFL74およびFGL細胞の2種類のリンパ肉腫細胞において無制限の状態でα鎖の発現が認められ、特にFGL細胞においては外因性のIL-2添加によりIL-2Rα鎖の発現のUP-regulationが観察された。
