| Research Abstract |
実験肝癌では変位増殖巣は肝細胞癌の重要な前駆病変とされており、先にこれら細胞の核DNA ploidyパターンが正常肝細胞のものとかなり異なることを明らかにした。細胞核DNA ploidyは細胞増殖能を反映するものとされるが細胞核小体も生理機能より核内DNA量と密接な関連を有するとされる。変位増殖巣細胞の核小体オルガナイザー(NOR)の解析をN-γ-fluoreny Lacetamide(FAA)を0.02%、20週間飼料投与された♂ACIラット肝に於いて行った。肝組織はH・E染色、NOR染色、抗BrdU、単クローン抗体免疫染色(殺前にBrdU投与)の3枚の連続切片として観察された。平均NOR数とBrdU標識率(%)は変異増殖に於いて好酸性型:1.78、1.82、明細胞型:1.45、1.77、好塩基性型:1.99、4.58、超塩基性型:2.94、8.02(非病変部:1.33、0.13、無処理肝:1.20、0.08)であり、NOR数とBrdU標識率の増殖巣各型間、増殖巣と非病変部ないし正常肝との相関性がDNA ploidyパターンのものと良く似ていることが分った。DNA ploidyパターンの組織切片のspectrophotometerによるもの、自由細胞でのflow cytometryによるものとの相違について検索した。ACI、F344、WistorラットにそれぞれFAAを投与し変異増殖巣を多数有する肝を作成、Collagenase潅流により自由細胞を得た。γ-glutamyltronspeptidase染色により変異増殖巣の出現率を確認した後、flowcytomethyにてDNA ploidyパターンを解析した(対照ラット肝との比較により変異増殖巣核DNA poidyパターンを推計した)。Flowcytometryによるものは2N、4N集積傾向ありspectrophotometerによるものに比しploidy低下を示した。人肝の肝細胞癌関連病変をfocal nodular hyperplasia,adenomatoid hyperplasia,partial nodular transformation,liver cell clysplasia,altered focus等に分類した(Altered focusは好酸性型と好塩基性型)。
