Outline of Annual Research Achievements |
We established the star maze behavioral protocol in the laboratory and successfully completed analyses of navigational selection in control cohorts of animals. We characterized a novel Cre recombinase expressing transgenic mouse line that provided genetic access to principle neurons of the retrosplenial cortex. DREADD-based pharmacogenetic silencing approaches failed to produce a quantifiable change in navigation behavior in these animals. In vivo recording was used to verify the action of the circuit manipulations. We found that the administration of the DREADD ligand, CNO, was effective in reducing the firing rate of a substantial fraction of the neurons we recorded in the retrosplenial cortex. The absence of a clear behavioral impact of this silencing on the navigational strategy selection in the experimental mice remains to be clarified with future work.
