Outline of Annual Research Achievements

申請者は、これまでバナジウム金属触媒、酸塩基複合有機分子触媒、及び金属・有機分子ハイブリッド触媒等の多機能不斉触媒の開発に成功している。これら触媒を活用することで、光学活性オキサヘリセンや連続キラル四置換炭素を含む複素環、軸性キラリティーを有するアルカロイドなど、ラセミ体として得るのも困難な複雑化合物の短工程合成を可能としている。これら反応を効率的に促進するには、触媒を構成する複数の活性化部位による反応基質の多点配向制御が重要であり、酵素的作用機序を持つ不斉触媒としての知見も数多く得ている。そこで本研究では、多機能触媒が促進する連続反応をさらに集積化し、既存法(バッチ法)では合成が困難な多置換・多官能性キラル骨格の触媒的かつエナンチオ選択的構築法を開拓する。触媒をリサイクル可能な固定化触媒へと導き、省資源・環境調和型不斉反応プロセスへと展開する。複雑な多成分連結反応をマイクロフロー合成に組み込むことで、生物活性天然物や医薬原料の供給に有効な実用的不斉分子変換法の確立を目指す。
本年度は 金属・有機分子ハイブリッド触媒が促進する不斉ヘテロカップリング反応と光学活性ビカルバゾールアルカロイド類のドミノ反応集積型ダイバージェント合成を主テーマに研究を行った。ヒドロキシカルバゾールの二量体であるビヒドロキシカルバゾールは、天然に広く存在し、抗菌活性や抗酸化作用など生物活性を示す。しかし、化学安定性の低いヒドロキシカルバゾールの網羅的かつエナンチオ選択的な合成法は確立されておらず、その絶対配置も未決定なものが多い。今回、二核バナジウム触媒とバナジウム有機分子ハイブリッド触媒とを使い分けることで、ヒドロキシカルバゾールのホモ及びヘテロカップリング反応を制御できることを見出した。応用研究として、Sorazolon E2の初めての不斉全合成を達成した。

Reason

不斉ドミノ反応の集積化において、マイクロミキサーの反応条件探索に機械学習を活用したところ、1分以内にキラルなスピロオキシインドールを合成可能な反応条件を見出すことができた。本ドミノ反応で合成した化合物の中にはWntシグナル阻害活性を示すものもあり、医薬資源としての応用も期待される。

Strategy for Future Research Activity

2019年度も引き続き、金属・有機分子ハイブリッド触媒が促進する不斉ヘテロカップリング反応を基軸とする光学活性ビカルバゾールアルカロイド類のドミノ反応集積型ダイバージェント合成を遂行すると共に、フロー合成を念頭に多機能不斉触媒の固定化研究を開始する。
