Outline of Annual Research Achievements

本研究では、C4光合成の律速因子として同定されたCP12-3を研究対象として、(1)CP12-3の構造機能解析を実施した。それに加えて新学術領域の他の研究者と共同で(2)光合成関連酵素群の構造機能解析を行った。以下に詳細を述べる。
(1)については、フラベリアCP12-3の発現・精製、ならびにCP12-3の結合標的候補の酵素としてMDH, NADP-ME, PRK, GAPDHの発現・精製を試みた。CP12-3, MDH, NADP-MEについては大腸菌での発現・精製系構築することができた。さらに、これまで達成できてこなかったGAPDHについては、CP12-3と共発現することによって、大腸菌での発現・精製系を構築することができた。その後、CP12-3とGAPDHが相互作用することがわかったため、CP12-3とGAPDHの相互作用解析を実施した。その結果、CP12-3がGAPDHの非特異的な集合を抑制する可能性を見出だすことができた。さらに、CP12-3/GAPDH複合体、およびGAPDH単体の構造解析に取り組んだ結果、GAPDH単体の構造解析に成功した。通常、四量体で存在するGAPDHが二量体で存在することが明らかとなり、この結果からCP12-3の新しい機能、すなわちGAPDHの構造安定化に寄与する可能性が示された。
(2)については、ソルガムRubiscoのRbcSをイネに導入し、さらにCRISPR-Cas9で野生型イネ(Nipponbare)のRbcSをノックアウトしてハイブリッドRubiscoを作成したところ、酵素活性が1.9倍、光合成活性も1.2倍程度上昇することが分かった。その成果を最近論文として発表することができた(Mol Plant, 13, 1570, 2020)。現在は、この分子機構を元に、ソルガムとは異なるRbcS遺伝子を導入した改良型イネの研究を展開し、さらに詳細な分子機構がわかりつつある。
