ヘリオロドプシンは何を伝えているのか?

Outline of Annual Research Achievements

これまで地球上のロドプシンには、微生物に由来するタイプ1と動物に由来するタイプ2しか存在しないと考えられていた。ところが我々は2018年、全く新しいロドプシンを発見し、第三のロドプシンであるこのタンパク質群にヘリオロドプシンと名付けた。ヘリオロドプシンはイオンを輸送しないため、我々は光センサー機能を推測しているが実験的な証拠はない。ヘリオロドプシンが古細菌、真正細菌、藻類などの真核生物から巨大ウイルスにまで存在する事実は、普遍な光利用機構の存在を示唆している。果たしてそれがタンパク質間相互作用によるシグナル伝達なのか、あるいは全く別の機構が存在するのか、実験的に明らかにすることを目標とした。2020年度は以下の成果が得られた。
古細菌由来のヘリオロドプシンについて、前年度の野生型に続き、東大との共同研究によりE108D変異体の結晶構造解析に成功した。これにより、ヘリオロドプシンが示す特異な波長制御機構に関する構造的な示唆を得ることができた。
一方、全反射赤外分光を用いた解析により、亜鉛が特異的に結合することがわかった。亜鉛の結合は0.2 mMの解離定数であり、他の金属と異なり結合によりタンパク質骨格の変化を伴うため、その結合が特異的であることがわかる。この実験事実は、現在も未知であるヘリオロドプシンの機能に対して、亜鉛の結合が影響を与えることを示唆するものである。
また系統樹において、タイプ1ロドプシンとヘリオロドプシンの中間的な位置に存在するシゾロドプシンが内向きプロトンポンプの機能を持つことを明らかにした。
