Outline of Annual Research Achievements

一般的に相互作用の無いとされている双性イオン間の弱い相互作用について調べることを本研究の目的とする。特に、双性イオンの構造や、溶液の条件によって相互作用が変化するという点に着目する。例えば合成高分子の双性イオン高分子と、細胞膜最表面のホスホリルコリン基の相互作用を制御することで、細胞内への高分子の取り込みの促進や抑制を調節できるようになると期待される。
生体適合性高分子として知られているポリ（2-メタクリロイロキシエチルホスホリルコリン）（PMPC）の側鎖とは、逆の電荷の順番のコリンホスフェート（CP）基を側鎖結合したコリンホスフェートポリマー（PMCP）を制御ラジカル重合法により合成した。得られたPMCPについて、さまざまな方法でキャラクタリゼーションを行った。PMCPは純水中でポリマー鎖間の静電相互作用で会合することを確認した。PMCPの水溶液に塩を添加すると、ポリマー間の相互作用は遮蔽された。

Reason

今年度、コリンホスフェートポリマー（PMCP）を制御ラジカル重合法で合成した。重合の際の動力学的挙動について調べた結果、重合中の成長ラジカルの濃度が一定であることを確認した。また、得られたPMCPの分子量分布は、せまかったので、重合はリビング的に進行したと考えられる。得られたPMCPの純水中での動的光散乱および1H NMRのDOSY測定により求めた拡散係数から、流体力学的半径を見積もると、会合していた。この水溶液に食塩を添加すると、粒径の減少が観測されたため、会合のドライビングフォースは静電相互作用であることが示唆された。また、疎水的環境で蛍光極大波長が短波長シフトすることが知られている蛍光プローブである、フェニルナフチルアミン（PNA）を用いて、ポリマーによる疎水性ドメインの形成について調べた。PMPCおよびPMCPのどちらの場合も、高濃度の水溶液中で、疎水性ドメインを形成していることがわかった。複数の弱い相互作用がポリマー間に働くことで会合体を形成していることが示唆された。

Strategy for Future Research Activity

今後は、PMPCやPMCPをモデルとして、側鎖にリン酸基を含むモノマーと、側鎖に4級アンモニウム塩を含むモノマーのランダム共重合体であるポリアンホライトの作製を試みる。PMPCやPMCPのようなベタイン構造の高分子は、一つの側鎖にカチオンとアニオンの両方を含むが、一つの高分子鎖中にカチオンとアニオンをランダムに共重合体したポリアンホライトの場合も、タンパク質等との相互作用が抑制されることを確認する。また、ポリアンホライトの場合、カチオンとアニオンの比率を変えることで、トータルのポリマーの荷電状態を制御できるので、カチオン性やアニオン性のポリアンホライトを作製できる。このような電荷比のずれたポリアンホライトと、タンパク質や細胞との相互作用について調べることで、合成高分子と細胞間の弱い相互作用について評価する。
