Outline of Annual Research Achievements

植物は周期的に幹細胞群を新たにつくり、枝分かれを行う。ゼニゴケは、葉状体の先端に存在する頂端幹細胞を周期的に増幅して二叉分枝を行うが、その際の幹細胞の動態はよくわかっていなかった。植物ホルモン・オーキシンへの応答性が、幹細胞ゾーンでは低く、その周辺では高いことが示唆されたため、オーキシン応答マーカーを確立して、その動態を追う予定であったが期間内には確立できなかった。しかし、幹細胞ゾーンで特異的に発現する転写因子(S)と、オーキシン応答促進に関わる転写因子(A)の発現レポーターを用いて、ライブイメージング観察を行ったところ、分枝進行に伴ってAの発現が低い領域が拡大し、Sの発現はその領域中で揺らぎながら最終的に両端に固定される様子が観察された。よって、新しい幹細胞は元の幹細胞から直接生じるのではなく、低オーキシン応答性領域が拡大し、その両端に新たに形成されることが示唆された。
AとSの標的候補として前年度に同定した、分泌性ペプチドとその推定受容体の遺伝子の発現レポーター株を観察したところ、AとSの発現部位とよく合致する発現パターンが観察された。ペプチド遺伝子の誘導過剰発現株で発現誘導を行ったところ、幹細胞ゾーンが乱れるとともに、メリステム内で分化に向かう細胞がより早く分化すると考えられる表現型が見られた。よって、このペプチドを介した細胞間コミュニケーションが、オーキシン応答性制御と協調して幹細胞性を制御することが示唆され、それが幹細胞動態、およびオーキシン応答性の区画化に及ぼす影響をさらに解析することで、二叉分枝のより詳細なメカニズムの解明が期待される。
