Outline of Annual Research Achievements

多くの生体分子系は同種の色素をタンパク質内に含む。高速分光を用いた場合、同種分子間のエネルギーや電子伝達にはスペクトルでは判別がつかず、その反応経路の因子を明らかにすることはできない。そこで本研究は、三次元に分子を配列した生体分子内の、同種分子を含む励起状態、電荷分離状態の局所的な選択的観測を可能とする方法論を開発し、生体たんぱく質反応において未解明な部分を明らかにすることを目的とした。
結晶試料の吸光度の偏光依存性を示す描画手法に極座標プロットがある。この極座標プロットにより、三次元的に高度に配置された分子の局所的な吸収成分を、特性吸収波長と遷移モーメントの違いから議論することが出来る。結晶において複数のエネルギーの電子遷移が、複数のモーメントで存在する場合、偏光軸毎に吸収の異方性が変わる。この異方性値はそのダイナミクスとともに定量的に計測、解析することができる。したがって、信号の異方性値を解析することにより、電子移動反応、エネルギー移動反応を理解することができる。本研究では二年間でこの吸収の異方性解消を観測する、時間分解異方性解消実験手法を過渡吸収分光法に取り入れ、過渡吸光度の異方性の時間変化をスペクトルで計測し、その波長依存性を明らかにすることで、三次元構造を有するタンパク質内の局所的な分子を選択的に時間分解観測する装置を開発し、光合成エネルギー伝達系タンパク質結晶に応用した。
