着床前胚におけるストレス応答の分子機構と疾患発症基盤としての意義

Research Abstract

初期胚において、ミトコンドリアに局在するNAD依存性脱アセチル化酵素であるSirt3の機能低下がROS産生の亢進を介して胚発生を遅延または停止させること、その機序にp53が関与することを報告してきたが、p53+Sirt3ダブルノックアウトマウスを用いた検討から、p53とSirt3の欠損は胚発生の進行には互いに相反する効果を及ぼし、p53依存性経路の抑制が胚発生率を改善させる可能性を明らかにした。一方、成獣マウスの解析から、成長後の病態形成(腫瘍の発生)には相乗的に働くことが示唆された。また、初期発生においてはSirt3 mRNAの導入によりROS刺激による胚発生の抑制が緩和されること、ROSが関与すると考えられている排卵後の長時間培養による加齢性変化に対し、防御的に働くことなどが明らかになpた。さらに、加齢マウスからの卵子ではSirt3 mRNAレベルの低下しており、体外授精後の胚発生率が有意に低下することを確認した後、Sirt3 mRNAの顕微注入による効果を解析し、その初期発生率が改善することを見出した。Sirt3過剰発現の効果とその機序をさらに明らかにするため、Cre-loxP依存性のコンディショナルSirt3過剰発現マウスを作成し、臓器特異的Cre発現マウスを用いてその有効性を確認した。
現在卵子特異的高発現マウスの作成と解析を進めている。以上の結果から、Sirt3の導入または内因性Sirt3の発現誘導が胚発生においてストレス応答性を改善させる可能性が示され、高齢女性の生殖補助医療においてSirt3を標的とした老化卵子の人工受精率改善法の開発が期待された。
