| Research Abstract |
1) プレウロサイクロアジリジンのプローブ化
プレウロサイクロアジリジンはスギヒラタケ食中毒の原因物質として注目されており，当研究室では当該分子の改良合成法を確立した．この合成では，D-セリンから誘導されるガーナーエステルに過剰量のMeMgBrを作用させジメチル基の導入を行ったが，プレウロサイクロアジリジンのプローブ分子の合成では，ガーナーエステルへのアルキル基とメチル基の段階的な導入により，リンカーを有する前駆体を合成した．今後，これまでと同様に末端のアミノ基の導入はNs-strategyにより行い，アジリジン環は光延反応により構築することで蛍光プローブが合成可能である．合成したプローブ分子を用いてスギヒラタケ食中毒の原因究明に取り組むことができる．
2)MFPAのプローブ化
強力なグルタミン酸受容体アゴニスト活性を有するMFPA は，脳機能解明に貢献する分子ツールとして期待さ れており，申請者は，分子間C-H挿入反応を鍵段階としてMFPAの全合成を達成している．本合成法を基盤として合成中間体であるMFPA保護体の芳香環上メトキシ基のパラ位選択的なヨウ素化反応を行った後，得られた化合物のヨウ素原子を起点として，プロパルギルアルコールと園頭カップリングによりリンカー部分を導入し，アルコールをアジドへと変換することでプローブ前駆体を合成した．得られた前駆体と別途調整したTokyoGreen（TG）誘導体とを脱水縮合し，蛍光プローブ分子の合成を達成した．合成したプローブ分子は顕微鏡による解析により生細胞での動態の観察を 可能にするだけでなく，競合実験にも応用できる特徴があり，当該分野における研究の発展が期待される．
